Most plumbing work falls into a few kinds of job: leaks and burst pipes, blocked drains, water heater repair and replacement, fixture installation for kitchens and bathrooms, and re-piping an older house. Some plumbers also hold a gas license and can work on boilers and gas lines. Many do not, so it is worth asking.
Ask whether they are licensed in your state, and for the number. Ask whether the quote covers parts and labor or only the call-out. Ask who actually does the work, since some firms subcontract. For anything involving gas, ask specifically about a gas license, because a general plumbing license does not always cover it.
A burst pipe, a leak you cannot isolate, sewage backing up, or no hot water in freezing weather. If water is running and you cannot stop it, find your main shut-off valve first, then call.
